Cell culture is the technique that allows multicellular organisms' cells to be kept alive and proliferating outside the body — under controlled conditions of temperature, pH, CO₂, and sterility. It is the basis of diagnostic microbiology (blood cultures, urine cultures), biopharmaceutical production, oncology research, and cell therapies. This guide explains the essential equipment for establishing and maintaining a functional cell culture laboratory in Colombia.

1. The Cell Culture Equipment Ecosystem

🌡️
CO₂ Incubator
The most critical piece of equipment in the cell culture laboratory. It maintains a constant temperature of 37°C, a CO₂ concentration of 5% (to buffer the culture medium and maintain pH 7.4), and 95% relative humidity. Models with dry heat (140°C) or UV decontamination minimize the risk of fungal contamination. Examples: Thermo Scientific HERAcell 150i, Esco CCL-170B, BINDER CB 150.
→ 37°C · 5% CO₂ · 95% RH · Integrated decontamination
🛡️
Class II Biosafety Cabinet (BSC)
The Class II Type A2 biosafety cabinet is the most important containment equipment in the cell culture laboratory — it simultaneously protects the operator, the cell culture, and the environment. Vertical laminar flow with HEPA filter. NSF/ANSI 49 certified. Mandatory for working with Class 2 risk biological agents (most human cell lines). Requires semi-annual validation.

🔭
Inverted Microscope
The cell culture microscope has the objective below the stage — the opposite of a conventional microscope. This allows cells to be observed through the bottom of the culture flask (where they grow attached) without the need to transfer or manipulate them. Essential for assessing cell morphology, confluence, signs of contamination, and treatment effects.

🌀
Cell Centrifuge
Low-to-medium speed centrifuge (300–1000 × g) to separate cells from the culture medium, wash cells, and perform density gradients. The swing-out rotor centrifuge is preferred for the cell pellet — it produces a flatter pellet that is easier to resuspend. Typical capacity: 4–8 tubes of 15–50 mL.
→ 300–1000 × g · Swing-out rotor · Gentle centrifugation

2. Complementary Equipment for the Cell Culture Laboratory

❄️
Thermostatic Water Bath
For thawing cryopreserved cell vials at 37°C in 1–2 minutes and warming culture media before use. Precise temperature ±0.1°C.
🧊
Cryopreservation and Liquid Nitrogen
Liquid nitrogen tank (-196°C) for long-term storage of cell lines. Controlled freezing (at -1°C/minute) with DMSO as a cryoprotectant is the standard protocol.
💜
Cell Counter (Hemocytometer / Analyzer)
Hemocytometer (Neubauer chamber) for manual counting with trypan blue (viability). Automatic cell counters (Nexcelom Cellometer) with image analysis automate the count and measure viability simultaneously.

3. Biosafety Levels and Regulation in Colombia

Biosafety Levels (BSL) and What They Entail in Colombia Work with cell cultures in Colombia is regulated by the Ministry of Health's Biosafety Manual and the INS resolution on biological agents. Most established cell lines (HeLa, HEK293, Vero) are BSL-1 or BSL-2 level — they require a Class II biosafety cabinet and standard microbiological good practices. Work with primary human tumor cells or virally transformed lines falls under BSL-2 and requires approval from the institutional biosafety committee, laboratory registration with the INS, and certified biological waste disposal procedures.
What culture medium is used for most cell lines?
The most universal medium for mammalian cell lines is DMEM (Dulbecco's Modified Eagle Medium) supplemented with 10% fetal bovine serum (FBS) and antibiotics (penicillin/streptomycin). Specific lines may require specialized media: RPMI 1640 for lymphocytes and hematological lines, MEM for Vero cells and fibroblasts, Ham's F-12 for keratinocytes and epithelial cells. Modern serum-free media are gaining ground in research and biopharmaceutical production to eliminate FBS variability.
